He has worked on the modular strategies of synthetic biology for combinatorial construction of yeast metabolic system for natural product biosynthesis. One of his main researches is focused on developing a new strategy (M-PERL) to introduce DNA sequence libraries into multigene pathway. This strategy utilizes small double-strand DNA as regulatory linkers to assist assembly of multiple gene parts. The regulatory linker contains a central regulatory region and two outside homologous ends, which were alternatively homologous with two orthogonal gene parts ready for assembly. With this strategy, several yeast promoter libraries covering varied strengths were developed. Alternative libraries inserting 5, 20, 40 random basepairs into five different promoters are used to regulate a five-gene pathway for violacein synthesis. Each gene expression and pathway’s product profiles are significantly regulated. This strategy is highly efficient for one-step assembly and regulation of multigene pathways.
